How to define color accuracy of an image system

Hi,
I am working in image analysis but until now I have very little experience in color measurements. I need to measure the amount of purple in an image. Therefore it seems logical to first determine the accuracy of my camera system. So how do I know that the amount of purple measured in the image corresponds to the real amount in the object? I was thinking of using a "golden standard" like spectrophotometry. Anyone how has experience in this?

Yeah, I might know a little bit about color. I'm in charge of color measurement for several systems that produce products used by a few billion people. I also teach courses on color science and attend color conferences. Many in the company consider me an expert but I don't know everything about color and am always learning (as are the top experts in the world on color). An expert is someone who learns more and more about less and less, until eventually they know everything about nothing at all.
For color calibration, see my answer here: http://www.mathworks.com/matlabcentral/answers/105120#comment_178624.
For color segmentation, see some simple demos here in my File Exchange: http://www.mathworks.com/matlabcentral/fileexchange/?term=authorid%3A31862
