"This GUI will help the novice user get up to speed very quickly on using GUI-based applications. Everything is laid out in a very simple Step 1, Step 2, Step 3, etc. layout. It is a very good starting point for a typical image analysis application. MATLAB users tend to have a restricted view of what they call GUIs. MATLAB makes making an x,y plot so easy, that we don't call them GUIs. You may just want to look at the function plot.

Well maybe you should start with Doug Hull's online GUI tutorial videos.

Well, at the matlab command-line prompt type:
helpwin uicontrol
...from there browse through the help and documentation of how to build GUIs. Also look at the file-exchange contribution: http://www.mathworks.com/matlabcentral/fileexchange/24861-41-complete-gui-examples for a number of great illustrations. Also (if you're Spanish-literate) you might find: http://www.mathworks.com/matlabcentral/fileexchange/12122-handbook-of-graphical-user-interface-spanish. For more general information about the art of making good GUIs there are much in the interweb.
