Hi, I would ask if I'm using chan-vese segmentation methods, initiation mask must be square? if not can be any shape?

No the initial mask can be any shape although it is preferable to start with a relatively smooth shape such as a circle.
You can try different initial configurations using the function activecontour in Image Processing Toolbox that shipped with R2013a.
