Look to see where the nose is relative to the center of the face?

Unless, that is, the question is about determining left side from right side when one of them is mirror imaged? Looking around I see that there can be asymmetry in facial expressions during display of emotion that could potentially be analyzed.

Computer Vision Toolbox has a face detection function. It is based on viola-Jones, so if it is not accurate enough for your purposes then you could train an object detector using the toolbox.
