Incorporate images taken from different heights and form a 3D model

Hello,
I want to build a simple cell 3D model. Based on the reference, I could use the confocal microscope to take cell images (layer-based)from different heights (from bottom to the top) and incorporate them together.
Now I have taken the images, but I am wondering could Matlab import the images (.TIFF) and combine them as I just described? If yes, which function should I use?

I would try imread. If you want to do any calculation with them I would use something like:
for k=1:n
A(k,:,:)=double(imread(filename(k)));
end
