Sounds like it is a good approach. Have you tried anything yet? Which step are you having trouble with? Obviously I can't just hand over code to do the whole thing for you but can help you with small parts. You might want to study my Image Segmentation Tutorial: http://www.mathworks.com/matlabcentral/fileexchange/?term=authorid%3A31862. Where did you upload your images to?
