I'm sorry but unless you have a much more specific 'MATLAB-based' question, I'm guessing you won't get many responses here. If you could mention what algorithm you intend to use, and have specific doubts in it's implementation, you will definitely get more answers. In general, the first place to look for help in segmentation problems is to go through <http://www.mathworks.com/products/image/description5.html> and the related function reference.
