How can I save a plot on an image in a variable?

Hi all,
I have plotted some stuff on an image and now I would like to save it in a variable so I can apply more methods on the image.I looked online for an answer but I didn't find the answer that I am looking for.
Can anyone please help me Thanks in advance :)

Thank you very much for your help.It did work but the picture was noise so I had to make a median filter then remove the small objects to make the picture clear.
