i did this code to convert image to binary and make the digitization so i get it
if true
x=imread('mdb004.tif');
x=medfilt2(x);
x=im2bw(x,0.0706);
i2=imopen(i,strel('disk',50));
[bw, n]=bwlabel(i2,4);
i3=(bw==1);
image= x.*uint8(i8)
imshow(i3),figure, imshow(image)
end
