Not necessarily. It depends on the available drivers. I’ve successfully interfaced USB devices directly, since they created virtual serial ports.
It is also possible to use Arduino and Raspberry Pi with them.

You need the RTL-SDR support package:
which lists the following as "MathWorks Requirements":
- MATLAB
- Communications System Toolbox
- DSP System Toolbox
- Signal Processing Toolbox
- Simulink (Recommended)
