For starters you need to download and run my Image Segmentation Tutorial from my File Exchange. http://www.mathworks.com/matlabcentral/fileexchange/?term=authorid%3A31862 After that you can try some of the other things there like the color segmentation demos or the thresholding app. If you have questions about a specific image, ask back here in Answers.
