See FEX: Matt Fig's GUI examples for the programmatic control of GUIs. GUIDE is convenient, but not necessarily the best method to create GUIs.

No, Visual Studio is (or at least that's the easiest to use when designing the GUI and giving it a rich look with any control you could ever possibly want), but you'd need the Builder NE toolbox for that.
