Are you talking about the slightly raised surface or the hair?

Please annotate and upload a picture where you point, in red color, exactly to "my pixel THIN line" because I have no idea what you're talking about. Is there something there in that image that is expected to be just one pixel wide? I'm not seeing it, so please tell me where it is.

Try a bottom hat filter, imbothat().

Sorry for the confiusion guys, the THIN LINE is the line inside the red box, i try measure the width at point A, B , C and D and the exact length of the line inside the red box. So far, i try, enhance contrast, multiple threshold and use pixel to calculate it manually, but the pixel connectivity is not good.
