Refer to the explanation of pixelLabelIDs input argument for pixelLabelDatastore and make sure the following while loading the pixel Label images:

If your pixel label images are RGB images then for the pixelLabelIDs input argument mention the RGB triplet as m-by-3 matrix where m corresponds to the number of class names. Each row contains a 3-element vector representing the RGB pixel value to associate with each class name. Vectors must be unique. Use this format for pixel label data stored as RGB images.
If your pixel label images are grayscale then for the pixelLabelIDs input argument mention the m-by-1 matrix where m corresponds to the number of class names.
