Simulink External Mode Execution Error

I'm getting the error below when attempting to execute a Simulink program via Monitor & Tune on an Arduino Mega 2560.
Error occurred while executing External Mode MEX-file 'ext_comm':
Timed-out waiting for second connect response packet.
Unable to connect to the 'Arduino Mega 2560' target for ...
This error has only just started occuring (I have previously been able to run the same model on the same board) and I am unsure of the cause and possible ways to rectify.
Any support would be much appreciated.
