The row where you get an error does not exist in my fullfile function. What version of matlab are you using? Have you made local modifications in fullfile?

It looks like you've done hardly anything - just a single button. I'd just scrap it and start all over again. You've done so little that it's not worth trying to fix.

GUIDE is an integral part of MATLAB. If you can't even run it and create the most simple basic GUI, then that's an installation issue which means you get free telephone support from the Mathworks. You can call them for free and I suggest you do that.
